摘要 目的:观察JH42-1对人卵巢癌SKOV3细胞增殖抑制作用,探讨其对PCNA表达的影响。方法:以不同浓度JH42-1处理SKOV3细胞24、48、72 h,台盼蓝染色和MTT法检测细胞增殖抑制,免疫荧光细胞化学法检测PCNA表达。结果:不同浓度JH42-1均明显减少活细胞数(P<0.05)。JH42-1诱导细胞增殖抑制率呈剂量-时间依赖。处理48 h,PCNA明显降低(P<0.01)。结论:JH42-1抑制SKOV3细胞增殖,下调PCNA表达。 Objective To investigate the role of JH42-1 in inhibition of the proliferation of human ovarian cancer SKOV3 cells. Methods The SKOV3 cells were treated by JH42-1 for 24, 48, and 72 hours. The inhibition of cell proliferation was detected by MTY. Trypan blue assay was used to identify the survival cells. PCNA in the cells treated by JH42-1 was detected immunocytochemically. Results JH42-1 at different concentrations significantly decreased the number of the living cells (P 〈 0.05). ]H42-1 induced inhibition in a dose- and time-dependent manner, and it down-regulated PCNA expression. Conclusion JH42-1 can inhibit the proliferation of SKOV3 cells and down-regulate the expression of PCNA.
