重建Smad4基因对结肠癌细胞中Claudin-1、MMP-7 mRNA表达的影响 被引量:2

摘要 目的:研究Smad4基因对人结肠癌细胞中Claudin-1、MMP-7 mRNA表达的影响,进一步探讨Smad4发挥肿瘤抑制作用的机制。方法:将Smad4真核表达质粒以脂质体介导方法转染结肠癌SW480细胞,G418筛选稳定表达株,检测Smad4基因转染前后Wnt/β-catenin通路下游Claudin-1、MMP-7 mRNA的表达变化。结果:成功构建Smad4稳定表达的结肠癌细胞模型,转染组Claudin-1及MMP-7 mRNA表达显著降低。结论:Smad4能在分子水平抑制结肠癌细胞中Wnt/β-catenin信号通路下游Claudin-1、MMP-7的表达,提示Smad4除了参与经典的TGF-β信号通路外,还可能通过Wnt信号途径发挥肿瘤抑制作用。
