摘要
目的:观察靶向Xklp2靶蛋白(TPX2)和Aurora A在口腔鳞癌组织中的表达,探讨TPX2和Aurora A表达在口腔鳞癌治疗和预后中的意义。方法:选择口腔鳞癌标本61例,另选29例癌旁组织和61例正常口腔黏膜组织为对照组。应用免疫组织化学SP法检测TPX2和Aurora A蛋白的阳性表达,在显微镜下观察阳性细胞所占百分比,并进行结果判定。采用Spearman等级相关分析法分析TPX2和Aurora A的蛋白表达相关性。结果:免疫组织化学染色,TPX2蛋白在正常黏膜组织、癌旁组织和口腔鳞癌组织的阳性表达率依次增高,分别为4.9%(3/61)、51.7%(15/29)和86.9%(53/61),各组间比较差异均有统计学意义(P<0.05);Aurora A蛋白在正常黏膜组织、癌旁组织和口腔鳞癌组织的表达率依次增高,分别为3.3%(2/61)、44.8%(13/29)和72.1%(44/61),各组间比较差异均有统计学意义(P<0.05)。TPX2的蛋白表达水平与口腔鳞癌的淋巴结转移有密切关联(P<0.05)。随Aurora A表达水平的增高,口腔鳞癌的恶性程度和淋巴结转移的可能性也相对增加(P<0.05)。Spearman等级相关分析,在口腔鳞癌组织中TPX2与Aurora A蛋白的表达呈正相关关系(r=0.445,P<0.01)。结论:TPX2和Aurora A在口腔鳞癌的发生和转移过程中均具有重要的促进作用,且二者间也有相互促进的关系。
Objective To investigate the expressions of TPX2and Aurora A in oral squamous cell carcinoma(OSCC) tissue and explore the significances of TPX2and Aurora A expressions in the treatment and prognosis of oral squamous cell carcinoma.Methods 61cases of oral squamous cell carcinoma were chosen as experimental group,other 29cases of adjacent tissue of cancer and 61cases of normal oral mucosa tissue were chosen as control group.Immunohistochemical staining was used to detect the positive expressions of TPX2and Aurora A.The percentages of positive cells were observed under microscope.The correlation between TPX2and Aurora A protein expressions was analyzed with Spearman analysis.Results The results of immunohistochemical staining showed that the positive expression rates of TPX2protein in normal mucosa tissue,adjacent tissue of cancer and cancer tissue were 4.9%(3/61),51.7%(15/29),and 86.9%(53/61);and there were statistically significant difference among all groups(P0.05).The expression of TPX2protein was closely related to the lymph node metastasis of oral squamous cell carcinoma(P0.05).The positive expression rates of Aurora A protein in normal mucosa tissue,adjacent tissue of cancer and cancer tissue were 3.3%(2/61),44.8%(13/29),and 72.1%(44/61);and there were statistically significant difference among all groups(P 0.05).And with the increasing of Aurora A expression level,the malignant degrees and possibility of lymph node metastasis in oral squamous cell carcinomas were also increased,which had statistically significant difference among groups(P0.05).The TPX2protein expression in oral squamous cell carcinoma tissue had positive correlation with the Aurora A protein expression(r= 0.445,P0.01).Conclusion TPX2and Aurora A may play important roles in promoting the infiltration and metastasis of mucosal epithelium in oral squamous cell carcinoma,and the relationship between the two genes is collaboration and mutual promotion.
