乳腺癌组织中神经营养因子Neuritin的表达及其临床意义 被引量:4

摘要 目的:观察神经营养因子Neuritin蛋白和mRNA在人乳腺癌组织中的表达及其与临床病理特征的关系,探讨其在乳腺癌发生发展中的作用。方法:收集74例乳腺癌组织标本和12例正常乳腺组织标本,采用免疫组织化学和Western blotting法检测标本中Neuritin蛋白的阳性表达率和表达水平,利用RT-PCR法检测标本中Neuritin mRNA的表达水平。结果:Neuritin蛋白在正常乳腺组织中阳性表达率为25%,在乳腺癌组织中阳性表达率为70.62%,乳腺癌组织中Neuritin蛋白阳性表达率高于正常乳腺组织(P<0.05),且其在乳腺癌肿瘤细胞中定位于细胞核和细胞质。Neuritin蛋白的阳性表达率与乳腺癌淋巴结转移、TNM分期和ER/PR/HER2高阳性率有关联(P<0.05)。与正常乳腺组织比较,Ⅰ-Ⅱ期及Ⅲ-Ⅳ期乳腺癌组织中Neuritin蛋白和mRNA表达水平升高(P<0.05,P<0.01);与Ⅰ-Ⅱ期乳腺癌组织比较,Ⅲ-Ⅳ期乳腺癌组织中Neuritin蛋白和mRNA表达水平亦升高(P<0.05)。结论:Neuritin可能是促进乳腺癌发生发展的一个重要基因,也可能是治疗乳腺癌的关键靶向分子。 Objective To observe the expressions of neuritinrotrophic factor Neuritin protein and mRNA in human breast cancer tissue and the relationship between its expression and the clinicopathological features,and to explore its roles in the carcinogenesis and progression of breast cancer.Methods 74cases of breast cancer tissue samples and 12cases of normal breast tissue samples were collected.The expressions of Neuritin protein in samples were detected with immunohistochemical assay and Western blotting method;the expression levels of Neuritin mRNA in samples were detected by quantitative RT-PCR.Results The expression rate of Neuritin in normal breast tissue was 25%,and the positive expression rate of Neuritin in breast cancer tissue was 70.62%,which was higher than that in normal breast tissue(P0.05);Neuritin mainly located in the nucleus and cytoplasm.Meantime,there were positive correlations between the positive expression of Neuritin and lymph node metastasis,TNM stages and the positive rate of ER/PR/HER2.Compared with normal breast tissue,the expression levels of Neuritin protein and mRNA in stageⅠ-Ⅱ and Ⅲ-Ⅳ breast cancer tissue were increased(P0.05or P0.01);compared with stageⅠ-Ⅱ breast cancer tissue,the expresson levels of Neuritin protein and mRNA in stageⅢ-Ⅳ breast cancer tissue was significantly increased(P0.05).Conclusion Neuritin may be one of the genes to enhance the growth of breast cancer and may be a target for the treatment of breast cancer.
