不同处理方法对血浆组织型纤溶酶原激活剂活性的影响

摘要 目的比较不同处理方法对血浆组织型纤溶酶原激活剂(t-PA)活性的影响。方法检测3种不同的滤纸条(Whatman#1、Periopaper、国产滤纸)对血浆中t-PA酶活性的洗脱率,及观察收集血浆样本的Periopaper滤纸条在两种不同处理方法 (洗脱、非洗脱)、不同温度(-20℃、-80℃)、不同时间(1、2、4、12周)t-PA酶活性的保持情况。结果 3种滤纸条中,Periopaper对t-PA活性的洗脱率最高,Whatman#1次之,国产滤纸最差;在未洗脱状态下,-20℃和-80℃保存12周,t-PA活性几乎不受影响,而洗脱状态对t-PA活性影响较大。结论 Perio paper是理想的收集t-PA活性的滤纸条,未经洗脱并在-80℃状态下保存时t-PA活性能稳定保持3个月。 Objective To research the influence on the activity of tissue plasminogen activator (t-PA) in plasma sample with different disposal. Methods The elution efficiency of plasma t-PA activity were measured from three kinds of collection pa- per: whatman 1, periopaper, domestic filter paper. Periopapers soaked with plasma were divided into two groups. One group was treated eluting beforehand, another group was treated no eluting beforehand. Half samples of each group were stored at -- 20 ℃and the rest were stored at --80 ℃. The activity of t-PA of each sample were measured in 1 week, 2 weeks, 4 weeks, and 12 weeks. Results The results showed that periopaper had the highest elution efficiency of t-PA activity. In the condition of no eluting before hand, the t-PA activity of the samples stored in --20 ℃and --80 ℃ had no significant change within three months. Conclusion This study indicates that the periopaper is the ideal filter paper to collect GCF and the t-PA activity can re- tain at least three months in the condition of no eluting beforehand from strip paper with -80 ℃.
