Expression and Significance of Notch1 and MMP-2 in Human Triple-negative Breast Cancer

摘要 【目的】探讨三阴乳腺癌组织(TNBC)中Notchl、基质金属蛋白酶-2(MMP-2)表达及其意义。【方法】采用免疫组织化学S-P法检测56例TNBC组织和30例癌旁正常乳腺组织Notchl、MMP-2的表达情况并比较。【结果】TNBC组织中Notchl、MMP-2阳性表达定位于细胞质和(或)细胞核,其表达率分别为62.50%(35/56)、53.57%(30/56),明显高于癌旁正常组织(P〈0.01);56例TNBC组织中Notchl、MMP-2蛋白在组织学分级G3级阳性表达率高于G1+G2级(P〈0.05),Notchl、MMP-2的病理分期Ⅲ~Ⅳ期的阳性表达率高于I~Ⅱ期(P〈0.05),Notchl、MMp-2阳性表达与淋巴结转移有关(P〈0.05);相关性分析显示二者之间呈正相关(P〈O.05)。【结论】Notchl、MMP-2在TNBC中呈明显高表达,与TNBC的发生、发展和转移密切相关。 [ObjectivelTo explore the expression of Notchl and MMP-2 in human triple-negative breast cancer(TNBC) and its significance. [MethodslThe expressions o{ Notchl and MMP-2 in 56 TNBC tissues and 30 normal breast tissues adjacent to the cancer were examined and compared by using SP immunohistochemis- try method. [ResultsI The expressions of Notchl and MMP-2 in TNBC tissues were located in cytoplasm and/ or nucleus, and the positive expression rates were 562.50% (35/56) and 53.57% (30/56) respectively which were obviously higher than those in normal tissues adjacent to the cancer( P 〈0.05). In 56 TNBC tissues, the positive expressions of Notchl and MMP-2 in histological grade G~ were higher than those in G1+G2 ( P 〈 0.05). In addition, the positive expressions of Notch1 and MMP-2 in stage II- IV tumors were also higher than those in stage I-II tumors( P 〈0.05). Furthermore, the expressions of Notchl and MMP-2 were as- sociated with axillary lymph node metastasis( P 〈0.05). Correlative analysis showed a positive relation be- tween Notchl and MMP-2( P 〈0.05). [Conclusion] Notch1 and MMP-2 are highly expressed in TNBC, and their expressions are closely related with the occurrence, development and metastasis of TNBC.
