摘要
观察国产重组人白细胞介素 11(rhIL 11)对卡铂致猕猴血小板减少症的影响。成年猕猴每天静脉注射卡铂 (15mg/kg) ,连续 3天。第 3次注射卡铂后 18小时内开始皮下注射rhIL 11(每天为 5 0或 10 0 μg/kg) ,连续用药 14天。给药后定期检测外周血白细胞计数、网织血小板计数、血小板聚集功能和骨髓的集落形成和病理组织学变化等。结果发现 ,rhIL 11能明显提高注射卡铂猕猴的血小板计数最低值和缩短血小板计数少于 5 0 %药前值的持续时间 ,对白细胞和网织红细胞数的恢复也有一定程度的作用。给予rhIL 11后可见外周血网织血小板计数明显升高 ,但ADP诱导的血小板聚集功能无明显变化。rhIL 11组骨髓巨核系祖细胞和红系祖细胞集落形成增多 ,光镜下显示骨髓腔中粒系及红系细胞丰富和密集 ,巨核系细胞明显增多。上述结果表明 ,rhIL 11对卡铂致猕猴血小板减少症有明显的治疗作用 。
The effectiveness of rhIL 11 on thrombocytopenia induced by carboplatin in rhesus monkeys was investiga ted. Thrombocytopenia was induced in monkeys by i.v. administration of carboplatin at a dose of 15 mg·kg -1 ·d -1 for three consecutive days. rhIL 11(50 or 100?μg·kg -1 ·d -1 ) or Neumega (100?μg·kg -1 ·d -1 ) were administered s.c. for 14 days beginning one day following the final dose of carboplatin. The results showed that rhIL 11 significantly improved mean platelet nadirs and shortened the mean duration of platelet counts less than 50% of pre treatment values.Administration of rhIL 11 also resulted in moderate increase of the reticulated platelet, leukocyte and reticulocyte counts in peripheral blood and megakaryocytic and erythroid progenitors in bone marrow. rhIL 11 did not enhance ADP induced platelet aggregation.These results indicate that rhIL 11 has a potent thrombopoietic effect in vivo and could be an important agent to reduce the severity and duration of thrombocytopenia following chemotherapy.
