葡萄糖对体外培养hUVECs增殖以及凋亡影响

摘要 目的分析不同浓度的葡萄糖对体外培养的人脐静脉内皮细胞(hUVECs)增殖及凋亡的影响,并探讨其分子机制。方法体外培养hUVECs,与不同浓度的葡萄糖(28、42mmol/L)作用48h后,四甲基偶氮唑盐比色法测定hUVECs增殖率,流式细胞仪和Western Bloting技术分别检测细胞凋亡率及凋亡相关蛋白Bcl-2/Bax比值的变化。结果随着葡萄糖浓度的增高,hUVECs的增殖率逐渐降低,凋亡率逐渐增高,凋亡相关蛋白Bcl-2/Bax的比值逐渐降低,与空白对照组比较,差异均有显著性(F=11.08~249.49,q=5.38~27.59,P<0.05)。结论高浓度葡萄糖通过调节Bcl-2/Bax比值的变化,诱导hUVECs凋亡,同时抑制hUVECs的增殖。 Objective To analyze the influence of different concentrations of glucose on the proliferation and apoptosis of human umbilical vein endothelial cells (hUVECs) cultured in vitro and its molecular mechanism. Methods The hUVECs were cultured in different concentrations of glucose (28 or 42 retool/L) for 48 hrs. The growth rate of hUVECs was detected by MTT assay. The apoptosis of hUVECs and the Bcl-2/Bax ratio were detected by flow cytometry and western blot, respectively. Results With increasing of glucose concentration, the growth rate of hUVECs decreased gradually, and apoptosis increased, the ratio of Bcl- 2/Bax reduced, the differences being significant as compared with the blank control (F = 11.08- 249.49 ; q = 5.38- 27.59 ; P 〈 0.05). Conclusion High-concentration glucose induces apoptosis of hUVECs and, at the same time, inhibits the growth of hU- VECs through regulating the expression of Bcl-2/Bax protein.
