法舒地尔对大鼠心肌缺血再灌注后血管内皮功能影响 被引量:2

EFFECTS OF FASUDIL ON VASCULAR ENDOTHELIAL FUNCTION IN MYOCARDIAL ISCHEMIA-REPERFUSION RATS

摘要 目的探讨法舒地尔对大鼠心肌缺血再灌注后血管内皮功能的影响。方法选取雄性Wistar大鼠30只,随机分为假手术组、缺血再灌注组(IR组)、法舒地尔组,每组10只。法舒地尔组于缺血前1h腹腔注射法舒地尔10mg/kg;假手术组及IR组于缺血前1h腹腔注射等量的生理盐水。分别于再灌注后2、24h取血,采用酶联免疫吸附试验检测血清中一氧化氮(NO)、血管性血友病因子(VWF)的水平。并于再灌注24h后应用伊文蓝及TTC双染法确定心肌缺血面积。结果与假手术组相比,IR组再灌注2、24h后血清中NO含量显著下降,VWF活性显著升高(F=10.08~118.66,q=6.32~19.80,P<0.01)。与IR组相比,法舒地尔组再灌注2、24h后血清中NO含量明显升高,VWF活性显著下降(q=3.69~17.77,P<0.05)。与IR组相比,法舒地尔组再灌注24h后心肌缺血面积显著降低(t=6.01,P<0.05)。结论法舒地尔可以减轻心肌缺血再灌注后血管内皮细胞损伤,减少心肌缺血面积,对心肌具有保护作用。 Objective To investigate the effects of fasudil on vascular endothelial function (VEF) in myocardial ischemia- reperfusion (MIR) rats. Methods Thirty male Wistar rats were equally randomized to three groups as sham-operation, ischemi- a-reperfusion (IR) model, and/asudil. The rats in fasudil group were given intraperitoneal injection of fasudil (10 mg/kg) one hour prior to isehemia; those in sham-operation and IR groups were given same volume of normal saline. Blood samples were collected (2 and 24 hours after reperfusion) for detection of serum nitric oxide (NO) and von Willebrand factor (VWF) hy using ELISA. The ischemic area of cardiac muscle was determined by Evans-blue and TTC double staining-24 hours after reperfusion. Results At 2 and 24 hours after reperfusion, compared with the sham operation group, the serum NO level in IR group was significantly de- creased, and the VWF activity was significantly increased (F=10.08--118.66,q=6.32--19.80,P〈0.01); compared with IR, se- rum NO in fasudil group significantly elevated and VWF declined (q = 3.69-17.77, P 〈20.05). The ischemic area of m yoeardium in fasudil group much reduced as compared IR group at 24 hours after reperfusion (t =6.01,P〈0.05). Conclusion Fasudil reduces the damage to vascular endotheIial cell during myocardial ischemia-reperfusion, decreases the ischemic area, and protects myocar- dium.
