川芎嗪对人骨肉瘤MG-63细胞生长的抑制作用观察及机制探讨 被引量:3

摘要 目的探讨川芎嗪对人骨肉瘤MG-63细胞生长的抑制作用及可能机制。方法取对数生长期的人骨肉瘤MG-63细胞,随机分为川芎嗪低浓度组、高浓度组和对照组,川芎嗪低浓度组、高浓度组分别予3、30 mg/mL的川芎嗪,对照组加入完全培养液;48 h后采用四氮唑盐比色法检测MG-63细胞增殖抑制率;流式细胞术检测细胞凋亡率和细胞周期分布;Western blot法检测Bcl-2、CyclinD1及NF-κB p65蛋白表达水平。结果与对照组比较,川芎嗪低浓度组、高浓度组MG-63细胞增殖抑制率、凋亡率及G0/G1期细胞比例均明显升高,NF-κB p65及其靶基因Bcl-2、CyclinD1蛋白表达水平均明显降低,且川芎嗪高浓度组作用更明显;P均<0.05。结论川芎嗪对体外培养的MG-63细胞有增殖抑制作用,其机制可能与其促进MG-63细胞凋亡、诱导G0/G1期阻滞、抑制NF-κB及其靶基因的蛋白表达有关。 Objective To investigate the inhibitory effect and mechanism of ligustrazine on the growth of human osteosarcoma MG-63 cells. Methods Human osteosarcoma MG-63 cells of logarithmic growth phase were randomly divided into low concentration ligustrazine group, high concentration lignstrazine group and control group. Low concentration and high concentration ligustrazine groups were treated with ligustrazine of 3 and 30 mg/mL respectively, and the control group was treated with complete medium. After treatment for 48 h, the inhibition rate of proliferation in MG-63 ceils was detected by MTT assay. Apoptosis rate and cell cycle distribution were detected by flow cytometry. And the expression levels of NF-κB p65, Bcl-2 and CyclinD1 protein were detected by Western blotting. Results Compared with the control group, inhibition rate of proliferation, apoptosis rate and G0/G1 proportion of MG-63 cells in the low and high concentration ligustrazine groups were significantly increased; the protein expression of NF-κB p65 and its target gene Bcl-2 and CyclinD1 was significantly decreased; and the high concentration group was more effective ( all P 〈 0.05 ). Conclusion Ligustrazine can inhibit the proliferation of cultured MG-63 cells in vitro, whose mechanisms may be related to its effect on increasing apoptosis, inducing G0/G1 phase arrest and inhibiting protein expression of NF-κB and its target gene Bcl-2 and CyclinD1 in MG- 63 cells.
