摘要
为促进和加快降解菌的工业化生产,解决氯嘧磺隆残留药害问题,利用单因素筛选结合正交试验的方法,以氯嘧磺隆降解菌F31的菌体生长量(OD600)为测定指标,系统研究了不同碳源、氮源及无机盐的发酵结果。结果表明:单因素试验初步筛选出碳源葡萄糖,氮源蛋白胨和硫酸铵,无机盐KH2PO4、NaCl、MgSO4、FeSO4利于降解菌生长。在此基础上通过L18(37)的正交试验,确定了最优培养基配方为葡萄糖2%、蛋白胨1%、硫酸铵1%、KH2PO40.07%、NaCl 0.2%、MgSO40.07%、FeSO40.000 5%;最优配方培养基摇瓶降解能力试验表明:降解菌F31在新培养基中生长状态良好,降解能力优良,培养36h液相色谱条件下未检出氯嘧磺隆。
In order to promote and accelerate industrialization of degrading fungi, solve the residual phytotoxicity of chlorimuron-ethyl, with the single factor screening and orthogonal test, fermentation of different carbon sources,nitrogen sources and inorganic chemicals were systematically studied with index of degrading fungi growth(OD600 ). The results showed that in the single factor test, carbon source glucose,nitrogen source peptone and (NH4)2 SO4 ,inorganic salts K H2 PO4, NaC1, MgSO4 and FeSO4 were primarily screened out, which were benefit for the growth of the degrading fungi. Based on the single factor test,optimal carbon sources, nitrogen sources and inorganic chemicals were chosen to be component factors, and each of them was operated with orthogonal optimization test by three levels, thereby achieving optimum culture medium formula which was 1% glucose,2% peptone,l% (NH4)2 SO4,0.07% KH2PO4 ,0. 2% NaC1,0.07% MgSO4 and 0. 000 5% FeSO4. The best medium shake flask tests showed that F31 could grow well in the new medium and the degradation ability was strong,chlorimuron-ethyl with HPLC wasn't detected after 36 h.
