摘要
为探索斜纹夜蛾蛹畸形发育的生理机制,在室内将斜纹夜蛾幼虫饲养至蛹期,测定了斜纹夜蛾蛹的生物学参数,并采用双向电泳技术对蛹整体可溶蛋白质进行分析。结果表明:畸形蛹率达(18.8±1.2)%,其畸形蛹的发育历期明显短于正常蛹,为(9.5±0.7)d(P<0.01)。畸形蛹中雌蛹和雄蛹的体重也明显小于正常蛹(P<0.01)。在正常蛹中共检测到约147个蛋白质点,主要集中在分子量1.1×104至7.6×104、等电点3.23至9.80之间。在畸形蛹中共检测到149个蛋白质点,主要集中在分子量1.0×104至7.7×104、等电点3.61至9.61之间。在正常蛹中有45个特异表达蛋白质点,畸形蛹中有47个特异表达蛋白质点。
In order to explore the developmental mechanism of the malformed pupae in Spodoptera litura Fabricius, the larvae were raised in door till the pupal period. Partial biological parameters of S. litura pupae were determinated, and two-dimensional electrophoresis was used to analyze the component of soluble proteins in the malformed pupa. The results showed that the rate of malformed pupae was (18. 8依1.2)%, and the developmental duration of malformed pupae was (9. 5依0.7)days, shorter than that of normal pupae significantly (P〈0.01). The weights malformed pupae were lower than those of normal pupae significantly (P〈0.01). One hundred and forty seven protein spots were detected in normal pupa, mainly distributed in the range of 1. 1×10^4 -7. 6×10^4 of molecular weights and 3. 23-9.80 of isoelectric point(pI)values. 149 protein spots were detected in malformed pupa, mainly distributed in the range of 1. 0×10^4 -7. 7×10^4 of molecular weights and 3. 61-9.61 of pI values. There were 45 specific protein spots detected in normal pupa, while 47 were in malformed pupa.
