HPLC法测定黄连中盐酸小檗碱和盐酸药根碱

Determination of the Berberine Hydrochloride and Jatrorrhizine Hydrochlorid in Huanglian by HPLC

目的：建立黄连水提液中同时测定盐酸小檗碱和盐酸药根碱的高效液相色谱法。方法：加热回流法制备黄连药材水提液,反相高效液相色谱外标法同时测定盐酸小檗碱和盐酸药根碱的含量。结果：盐酸小檗碱和盐酸药根碱的的线性范围分别为：7.5~243.0μg/mL和0.78~34.2μg/mL,平均回收率分别为99.3%和100.1%供试。药材中盐酸小檗碱和盐酸药根碱的含量为21.3 mg/g和9.6 mg/g。结论：该方法能用于黄连药材中盐酸小檗碱和盐酸药根碱的含量测定。

Objective： to establish the method of HPLC to detect the berberine hydrochloride and jatrorrhizine hydrochloride at the same time in Huanglian.Preparation of aqueous extract of Rhizoma Coptidis heating reflux method was used to get the liquid of Huanglian,and the reversed-phase high performance liquid chromatographic method was used to determine the berberine hydrochloride and jatrorrhizine hydrochloride at the same time.Results the linear range of berberine hydrochloride and jatrorrhizine are： 7.5~243.0 μg/mL and 0.78~34.2 μg/mL,the average recovery rate was 99.3 % and 100.1 %.Respectively for the test material of berberine hydrochloride and jatrorrhizine hydrochloride for 21.3 mg/g and 9.6 mg/g.Conclusion the method can be used to determine the content of the berberine hydrochloride and jatrorrhizine hydrochlorid in Huanglian.
