不同种、株鸡球虫DNA多态性的比较研究 被引量:9

DIFFERENTIATION OF EIMERIA SPECIES(INTER AND INTRA SPECIES) BY THE RANDOM AMPLIFICATION OF POLYMORPHIC DNA

摘要 :对鸡的 4种球虫 (E tenella、E acervulina、E brunetti、E maxima)基因组DNA进行RAPD(RandomamplifiedpolymorphicDNA)比较分析 ,发现多数引物能使不同种球虫产生完全不同的谱带 ,因此证明RAPD技术完全可用于同宿主艾美耳球虫虫种的分类鉴定。并应用RAPD技术对E tenella早熟株、鸡胚株、亲本毒株和E acervulina早熟株和毒株的基因组DNA进行多态性比较研究 ,发现本研究选育的E tenella早熟株与亲本毒株间DNA多态性存在着差异 ,证实该早熟株已发生了遗传变异 ,但变异程度不大 ;而鸡胚株与毒株的差异程度比前者大。而且不同来源E acervulina 2株球虫间的变异程度大于E tenella各虫株间的变异 ,即不同来源的E acervulina各虫株间的相似性小于E tenella各虫株间的相似性 ,表明不同球虫其种内变异程度不一致。 The genomic DNA of E tenella、E acervulina、E brunettiandE maximawere analyzed by RAPD(Random amplified polymorphic DNA) Species specific DNA fragment mobility patterns were observed in most cases In the majority of assays conducted,the Eimeria species could be easily differentiated It seemed likely that RAPD assays,using appropriate primers,could distinguish most Eimeria species from one another The genomic DNA of the precocious line,parental line and embryo adapted line of E tenella and the precocious line and the virulent line of E.acervulina were analyzed by RAPD Genomic fingerprints generated by RAPD revealed a high degree of similarity(SI)between the lines of E tenellaand the lines of E acervulina The results showed that the similarity(SI=0 9240~0 9753)between E tenella lines was higher than that(SI=0 8169)between E acervulina lines It was suggested that the intra specific variance between the lines of different virulence,geography,was not consistent The results of this study demonstrated that the line specific fragment of Eimeria lines can be amplified by the RAPD technique,and the RAPD method may be an extremely useful tool for studies of the genetic organization of coccidial parasites Polymorphisms detected by RAPD PCR might facilitate the selection of molecular markers and study of the relationship between different lines
