摘要 目的:初步探讨DSS对去卵巢大鼠脂代谢的抑制机制是否与瘦素的调节有关。方法:选择清洁级SD雌性大鼠60只,体质量160-220 g,随机分成6组,①假手术组(正常对照组),②卵巢切除组(模型组),③卵巢切除+戊酸雌二醇组(阳性药物对照组),④卵巢切除+DSS高剂量组(100μg/kg),⑤卵巢切除+DSS中剂量组(50μg/kg),⑥卵巢切除+DSS低剂量组(25μg/kg)。术后1周连续阴道涂片检查10天后,①②组大鼠每天灌胃同体积生理盐水;③组大鼠每天一次戊酸雌二醇片剂水溶悬液灌胃,剂量为80 mg/kg;④⑤⑥组分别以高、中、低剂量的DSS溶液灌胃,1次/天。连续给药6周,每周一次测量体质量及体长,计算Lee's指数;麻醉后分离血清,分别测血清中Leptin含量及血清内HDL、LDL、TG、CHO的含量。结果:与模型组比较,DSS可降低Lee's指数,降低血清内CHO、TG、LDL含量,升高血清中HDL以及Leptin含量,差异有统计学意义。结论:DSS可减轻大鼠去卵巢后雌激素缺乏所致的肥胖,并可能通过升高血清中瘦素含量对大鼠脂代谢起调节作用。 Objective:To explore possible mechanism of the suppressive effects of 3'-daidzein sulfonate (DSS)on lipid me- tabolism in ovarieetomized (Ovx) rats. Methods : Sixty healthy female SD rats with weight of 160 - 220 g were respective- ly divided into the six groups randomly:sham-operated group (saline) , ovariectomized group (Ovx + saline), estradiol va- 1crate group( Ovx + 0.8 mg/kg) and DSS groups( Ovx + 100/50/25 μg/kg). Every week ,the weight and stem length were measured. All rats were treated for 6 weeks, and serum samples were collected to determine the high density lipoprotein (HDL) ,low or very low density lipoprotein (LDL/VLDL) ,triglycerides (TG) ,or cholesterol (CHO). And the density of leptin or visfatin in serum is detected by radio-immunity. Results:It showed that DSS significantly decreased Lee's index, and increased HDL,but reduced LDL/VLDL, TG, or CHO in serum. These findings indicated that DSS was effective in suppressing the obesity in ovariectomized rats. More specifically, leptin was decreased in serum from the rats by DSS, but increased by estradiol valerate. Conclusion: It is possible that DSS will maintain an efficient level of circulating leptin, thus preventing them in sensitivity to regulate the lipid metabolism and weight gain. It may represent a potential alternative therapy in the treatment of obesity abnormality in ovarian hormone-deficient women.
