色谱-质谱联用法测定人体内血浆中头孢克肟含量

Determination of Cefixime in Human Plasma by GC-MS

目的探讨色谱-质谱联用法测定人体内血浆中头孢克肟的含量。方法应用岛津气相色谱-质谱联用仪测定受试者体内血浆中头孢克肟含量,色谱柱为Rtx.5MS毛细管柱,流速为1.0 mL/min,质谱条件离子源温度200℃,EI电离方式。结果方法最低检测限为0.5μg/L,标准曲线方程:Y=2.4870X+0.3578,r=0.9978,精密度较高,回收率在80.0%左右,平均误差均<8%。结论色谱-质谱联用法测定人体内血浆中头孢克肟含量具有精密度好和回收率高等诸多特点,符合方法学的要求。

Objective To explore the determination of the content of cefixime in human plasma by gas chromatography-mass spectrometry.Methods SHIMADZU gas chromatography-mass spectrometry was used to determine the content of cefixime in human plasma by capillary column,Rtx.5MS column,flow rate 1 mL / min,mass spectrometry conditions of ion source temperature 200 ℃,EI ionization mode.Results The method detection limit was 0.5 μg /L,standard curve equation： Y = 2.4870X ＋ 0.3578 r = 0.9978,high precision,recovery rate was about 80.0%,RSD was less than 8%.Conclusion Gas chromatography-mass spectrometry determination of human plasma cefixime content has many characteristics such as good precision and high recovery rate,well meeting methodological requirements.
