摘要
探索通过生物信息学方法预测黄瓜绿斑驳花叶病毒(cucumber green mottle mosaic virus,CGMMV)的抗原表位,为基于抗原表位策略制备CGMMV抗血清打下基础。利用MEGA软件中的邻近算法对CGMMV进行系统进化分析,结果显示,不同CGMMV分离物可分为2个类群,不同分离物间未见明显的寄主相关性和地理相关性。应用不同生物信息学软件对CGMMV外壳蛋白不同区域的抗原指数、蛋白二级结构进行分析,结合外壳蛋白三级结构预测结果综合分析,CGMMV外壳蛋白抗原表位区可能位于N端3-15、80-90、90-100位氨基酸残基附近。
In the study,the epitopes in coat protein of cucumber green mottle mosaic virus (CGMMV) were predicted by several algorithms in some softwares.It was in favor of producing antiserum of CGMMV on the basis of the epitopes technique.Based on the nucleotide sequences of coat protein (CP) gene of different CGMMV isolates,phylogenetie analysis with the neighbor-joining algorithm of MEGA software showed that all isolates of CGMMV fell into 2 clades.The clades were not related with the geological origins and hosts of CGMMV.Some characteristics of CP,such as antigenic index and protein secondary structure,were analyzed with the algorithms in different softwares.Combined with predicted tertiary structure,the predicted epitopes were located in CP of amino acid position 3-15,80-90,and 90-100,respectively.
