枯萎病植物疫苗工程菌FJAT-9290生物学特性研究 被引量:1

摘要 以枯萎病植物疫苗工程菌(尖孢镰刀菌非致病性菌株FJAT-9290)为研究对象,以致病性菌株FJAT-282为对照,探索培养时间、碳氮源对2个菌株生长的影响差异,分析非致病性尖孢镰刀菌株FJAT-9290的生物学特性。结果表明:培养过程中菌株FJAT-9290与致病性菌株FJAT-282在菌丝干重和发酵液的pH值方面无显著差异;而产孢量在第6d时达11.47×106 cfu·mL-1,显著低于菌株FJAT-282的108.33×106 cfu·mL-1;菌株FJAT-9290的还原糖含量较菌株FJAT-282下降得快。碳源的影响表明:淀粉利于菌株FJAT-9290菌丝生长,葡萄糖利于其产孢;木糖不利于菌株FJAT-282菌丝的生长,果糖利于其产孢。氮源的影响表明:有机氮为氮源时菌丝生长速率快,铵盐既不利于菌丝的生长,也不利于产孢。氮源对菌落形态的影响比碳源大。 The biological characterlstms oi plant , 9290) were investigated by compare the effects of incubation time, and nutritional conditions on its growth with a pathogenic strain as a control, The results showed that there was no significant difference of the mycelium dry weight between strain FJAT-9290 with the pathogenic strain FJAT-282, the sporulation of strain FJAT-9290 was 1.47X106 cfu mL-1 on the 6th day, while the strain FJAT-282 is 108.33 X 10^6 cfu mL-1 , and changes of pH values had no significant effect on the culture growth. Furthermore, the concentration of reducing sugar in culture filtrate of strain FJAT-9290 decreased faster than strain FJAT-282 ; starch was conducive to the growth of mycelium of strain FJAT-9290, glucose was conducive to its sporulation, xylose was not conducive to the growth of mycelium of strain FJAT-282, fructose was conducive to its sporulation; organic nitrogen was benefit to the growth of mycelium, ammonium salt was not conducive to the growth of hypha, but also to the sporulation. In addition, nitrogen sources exercised a greater influence on colony morphology.
