二陈汤合桃红四物汤防治大鼠非酒精性脂肪肝的实验研究

Study of Erchen Decoction Plus Taohong Siwu Decoction for Preventing and Treating Non-alcoholic Fatty Liver Disease in Rats

【目的】探讨二陈汤合桃红四物汤对非酒精性脂肪肝大鼠血脂代谢及对肝脏病理的影响。【方法】将SD大鼠随机分为空白组,模型组,易善复组(20 mg/kg),中药高、中、低剂量组(40、20、10 g/kg),高脂饲料喂养4周复制脂肪肝模型后,各给药组给予药物灌胃4周并继续给予高脂饲料。第57天采血检测转氨酶和血脂4项,采用酶联免疫吸附法(ELISA)检测脂联素(APN)水平,光镜下观察肝组织病理改变,采用扫描电镜观察肝窦结构。【结果】二陈汤合桃红四物汤高剂量组能降低脂肪肝模型大鼠血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)含量,升高高密度脂蛋白(HDL-C)和血清APN的含量,其肝组织形态恢复趋向正常,中剂量组作用稍弱,低剂量组效果不明显。【结论】二陈汤合桃红四物汤可调节脂肪肝模型大鼠血脂水平并改善肝脏组织的病理变化,能在一定程度上防治脂肪肝。

Objective To observe the effect of Erchen decoction plus Taohong Siwu decoction （ED-TSD） on blood lipid metabolism and hepatic pathology in rats with non-alcoholic fatty liver disease （NAFLD） . Methods SD rats were randomized into blank group, model group, Essentiale N group （20 mg/kg）, and high-, middle- and low-dose ED-TSD groups （in the dosage of 40, 20, 10 g/kg, respectively ） . The rat model of NAFLD was established by feeding with high fat diet for 8 weeks and the rats in Essentiale N group and ED- TSD groups were given medicine from the 4th week. On day 57, serum aspartate aminotransferase （AST） , serum alanine aminotransferase （ALT） and four blood lipid indexes including total cholesterol （TC）, triglyc- eride （TG）, low density lipoprotein cholesterol （LDL-C） and high density lipoprotein cholesterol （HDL-C） were determined. Adiponectin （APN） level was detected by enzyme-linked immunosorbent assay （ELISA） .The pathological features of hepatic tissue section were observed under light microscope, and the uhrastructure of hepatic sinusoid were observed under scanning electron microscope. Results In high-dose ED-TSD group, serum levels of AST, ALT, TC, TG and LDL-C were decreased, while those of HDL-C and adiponectin were increased. Besides, hepatic pathological tissue become normal. Conclusion ED-TSD can regulate blood lipid levels and improve hepatic pathological changes, thus it can prevent and treat NAFLD.
