摘要 目的 研制NOVA16全自动急诊生化分析仪试剂包中A、R、H液。方法 根据电极法测定血浆 (或全血 )中Na+ 、K+ 、Cl-、TCO2 、Glu、Urea、Cr等物质浓度 (活度 )和红细胞压积 (Hematocrit,Hct)的原理及标准液配制的有关要求 ,配制类似血浆水的A液、CO2 释放液H及参比液R。结果 自配A、R、H液与原装试剂的主要理化指标 :pH、渗透压、电极斜率、线性范围及试剂的稳定性等方面均一致。自配试剂的批内、批间精密度符合要求 ,与原装试剂之间的相关性好 (r =0 95 0~ 1 0 0 0 ) ,自配试剂所测结果批内、批间稳定性好 (P >0 0 5 ) ,长期使用对电极无损害。结论 自配试剂的准确性、精密度符合临床要求 ,提示自配A、R、H液可代替原装试剂A、R、H液。 Objective To develop solution A、 R、 H in NOVA16 analyzer reagent kit. Methods On the principle of electrode method for detecting Na+、 K+、 Cl-、 TCO 2、 Urea、 Cr、 Glu、 Hct and under the regulation of preparing standard solution, internal standard solution A、 similar to the constituents of plasma、 CO 2 releasing solution H and referance solution R were prepared. Results The main biochemical indicators such as pH, osmole, electrode slope and linear scope between self-made solution A、 R 、H and the primary reagent were in good accordance. The accuracy of results with self-made reagent was correlated well with those of the primary reagent (r=0950~100). There was good stability in the same lot (P>005) or among different lots (P>005) after six months and no damage to the electrodes was observed. Conclusion The accuracy of self-made reagent meets the qualifications of clinical application, and it can be used for the substitute of the primary solution A、 R、 H.
