摘要
本文采用双酶水解-高效液相色谱法,建立了一种肉松中淀粉含量的测定方法。研究结果表明,在选定条件下,α-淀粉酶和淀粉葡萄糖苷酶可将肉松中淀粉专一彻底水解成葡萄糖,采用高效液相色谱法测定样品中葡萄糖含量,在0.5—50.0mg/mL浓度范围内呈现良好的线性关系,线性相关系数为0.9999。采用不同浓度加标所得回收率为93.7%-99.8%,方法检出限为0.36%,定量限为1.2%,相对标准偏差(RSD)均小于5%。
A method for determination of starch in dried meat floss has been developed by double enzymolysis-High Performance Liquid Chromatography. In this study, starch in dried meat floss was divided into glucose by the enzymolysis of α -Amylase and Amyloglucosidase while the glucose was measured by HPLC. The calibration curve of glucose showed good linearity in range of 0.5 ~ 50.0 mg / mL with correlation of 0.9999. The detection limit of method was 0.36 % (S/N = 3), and the limited of quantification was 1.2% (S/N = 10).The average recoveries were in the range of 93.7%-99.8% with three different spiked levels, and the relative standard derivations were lower than 5%.
