反流物对食管组织内细胞增殖相关基因表达的影响 被引量:9

EFFECTS OF GASTRIC AND DUODENAL REFLUX ON THE EXPESSION OF CELL PROLIFERATION-RELATED GENES IN ESOPHAGEAL MUCOSA

摘要 用不同手术方式制作单纯胃食管反流 (G组 )、单纯十二指肠食管反流 (D组 )、十二指肠胃混合食管反流 (DG组 )及无反流对照组 (C组 )动物模型 ,于术后不同时期取出食管 ,用免疫组化法检测p16、PCNA、CyclinD1、CDK4 等基因的表达。结果显示 :D组、DG组大鼠食管组织中PCNA、CyclinD1、CDK4 基因表达显著高于G组 ,并随反流时间延长而明显增强 ,反流组与C组相比均有显著差异 ;各组不同时期p16表达相似。免疫组化结果与组织学改变高度一致。说明胃及十二指肠反流可能通过改变细胞周期调控基因的表达而致食管上皮增生 ,促进反流性食管炎及其并发症的发生 ,其中十二指肠内容物的作用更强。 To investigate the effects on expression of cell proliferation-related genes in esophageal mucosa of different reflux,we made three kinds of rat model (gastroesophageal reflux(G), duodenoesophageal reflux(D) and duodenogastroesophageal reflux(DG)) and control group(C) by operations. Their esophagi were taken at 1,2,4-week after operation and the expression of p16,PCNA,Cyclin D 1,CDK 4 were measured by immunohistochemical studies.The results showed that the expressions of PCNA,Cyclin D 1,CDK 4 increased in D and DG groups and were more significant than those in G group.Those in G group were more significant than those in C group. The expressions of p16 in each group were similar.We concluded that both of gastric juice and duodenal contents can make epithelial hyperplasia on rats esophagi by changing the expressions of cell proliferation -related genes.The effect of duodenal contents is stronger.
