TLR4在重症扩张型心肌病发病机制中的作用

摘要 目的通过观察扩张型心肌病(Dilated cardiomyopathy,DCM)患者外周血单核细胞(peripheral blood mononuclear cells,PBMCs)Toll样受体4(Toll—like receptor 4,TLR4)的表达情况,初步探讨TLR4在重症扩张性心肌病发病机制中的作用,以期发现治疗DCM的新思路。方法应用流式细胞术检测两组DCM患者及健康对照组外周血单核细胞TLR4的表达水平;用ELISA法测定三组血清中IL-6、hs—CRP的水平。结果DCM患者外周血单核细胞TLR4的表达明显高于健康人群(30.12±3.36),(36.27±1.82)vs(21.65±3.58),(P〈0.05),且其升高的程度与心衰的程度相关,与患者左室射血分数的值呈负相关。结论TLR4的表达与DCM的发病机制有关,且与心功能衰竭程度有关。 Objective By observing the expression of Toll-like receptor 4 ( TLR4 ) in peripheral blood mononuclear cells ( PBMCs ) ofpatiems with Dilated cardiomyopathy ( DCM ) , to explor the role of TLR4's effect in the pathogenesis of DCM, in order tofind new ideas for the treatment of DCM. Methods Detected TLR4 expression level by flow cytometry in PBMCs of the two DCM patients and healthy control group; the IL-6 and hs-CRP levels of three groups serum were measured by ELISA. Results The expression of TLR4 in PBMCs of patients with DCM was significantly higher than the healthy control group ( 30.12 ± 3.36, 36.27 × 1.82 ) vs ( 21.65 ± 3.58 ) , P〈0.05, The increased degree of the expression of TLR4 was related to the degree of heart failure. Conclusion The expression of TLR4 is related to the pathogenesis ofDCM, and it was related to the degree of heart failure.
