摘要 目的 建立一种快速测定体外培养表皮细胞生长增殖的方法.方法 利用MTT只被活细胞还原成紫色颗粒的特性,通过比色分析测定吸光度值,用于检测细胞的数量与活性;并与3H-TdR掺入法进行比较.结果 表皮细胞量在0.125~2.0×105细胞/孔时与所测定的吸光度值呈一良好线性关系(r=0.9689).用MTT法测定人表皮细胞在二种不同培养基中生长增殖与3H-TdR掺入法所测结果意义一致.结论 MTT法用于测定表皮细胞的生长增殖简便、迅速。 Objective This study was to devolop a new method that can rapidly measure the growth and survival of human keratinocyte HK. Method The colorimetric assay of MTT for living cells was utilized to determine the growth and Survival of human keratinocyte because MTT was only modified to a dark blue formazan product by living cell.The optical densityODvalues were read on a scanning multiwell spectrophotometerELISA reader. Results There was well linear relation between the optical density values and the quantity of HK at 0.125~2.0×105cells/well.The meaning of MTT method was similar to that of 3H-TdR for measuring proliferation of HK in two different kind of serum-free medium. Conclusion MTT method is convenientrapid and reliable for measuring the proliferation of HK it is worth spreading
