微生物谷氨酰胺转胺酶(MTGase)的分子生物学研究进展 被引量:2

摘要 谷氨酰胺转胺酶具有催化蛋白质分子内和分子间的交联,蛋白质和氨基酸之间的连接以及蛋白质分子内谷氨酰胺基水解的功能,被广泛应用于食品、医药及纺织等领域。微生物来源的谷氨酰胺转胺酶具有非Ca2+依赖性和底物特异性低等特点,在研究开发中更为引人注目。本研究综述了微生物谷氨酰胺转胺酶基因表达研究方面的进展,归纳了微生物谷氨酰胺转胺酶的基因序列特点及酶学特性,分析了该酶的结构与功能之间的关系,指出基因工程技术生产谷氨酰胺转胺酶的优势及存在的问题。认为应该进一步加强谷氨酰胺转胺酶的表达策略研究以及分子改造研究,以期提高该酶的产量、催化活力及热稳定性等。 Transglutaminase can generate covalent cross-links between protein molecules or intramolecular covalent cross-links to change functional properties of proteins, and is applied in industries of food processing, medicine, cosmetic and textile. Microbial transglutaminase has aroused much more attention because of its calcium-independent and broader substrate specificity. This article summarized the gene expression of microbial transglutaminase, reviewed the gene characteristics and enzymatic characteristics, analyzed the relationship between crystal structures and function, and introduced the advantages and problems of microbial transglutaminase production using genetic engineering technique. In order to improve the yield of the enzyme, the activity and thermo-stability, the suggestions about expression strategies and molecular modification of microbial transglutaminase were also provided.
