摘要
目的 研究酪氨酸激酶 (PTK)在类风湿关节炎 (rheumatoidarthritis,RA)成纤维样滑膜细胞 (fibroblast likesynoviocyte ,FLS)异常增生与转化特性中的作用。 方法 原代培养类风湿关节炎成纤维样滑膜细胞。应用形态观察、细胞计数、3H TdR掺入、流式细胞仪检测和双层琼脂培养法探讨染料木黄酮 ,特异性PTK抑制剂对于RAFLS细胞生长状态、数目、DNA合成、细胞周期及其非贴壁生长情况下细胞克隆形成数目的影响。结果 Genistein呈剂量依赖性抑制RAFLS的数目增长、3H TdR掺入和非贴壁生长的克隆形成数目 ;其生长抑制作用在细胞周期中G1期限制点。结论 酪氨酸激酶在RAFLS细胞异常增生与转化表现中发挥关键作用。
Objective To study the role of protein tyrosine kinase (PTK) in abnormal proliferation and transformation phenotype of rheumatoid arthritis fibroblast-like synoviocyte (RA FLS).Methods RA FLS was primarily cultured.Genistein,a protein tyrosine kinase inhibitor,was used to elucidate the effect of PTK on RA FLS morphology,number,DNA synthysis,cell cycle and colony numbers under anchorage-independent growth with microscope,cell counts, 3H-TdR incorporation,flow cytometry and LMP agarose culture method.Results Genistein had an inhibitory effect on RA FLS cell number, 3H-TdR incorporation and colony numbers under anchorage-independent growth in dose-dependent manner;the inhibition on RA FLS cell cycle was at G 1 restriction point.Conclusion PTK has a key role in abnormal proliferation and transformation of RA FLS.
