摘要
瞬时受体电位melastatin8(transient receptor potential melastatin8,TRPM8)蛋白是瞬时受体电位(transientreceptor potential,TRP)蛋白超家族中的一员,负责调控细胞内游离钙离子的动态平衡,在细胞增殖、凋亡以及分化中起着重要的作用。TRPM8最初作为一种前列腺特异性蛋白被克隆出来,之后发现其在肺癌、乳腺癌、结肠癌以及皮肤来源的恶性肿瘤中均有表达。最新研究发现,TRPM8不仅能感知冷知觉,还可作为前列腺癌诊断和治疗的新靶点。本文对TRPM8的结构、功能、定位、调控及其在前列腺癌发生和发展中的作用研究进行回顾,并对其在前列腺癌诊断和治疗中的应用前景进行展望。
As one of the members of superfamily of TRP (transient receptor potential),TRPM8 (transient receptor potential melastatin 8) regulates the intracellular free calcium ion homeostasis and plays an important role in proliferation,apoptosis,and differentiation of cells.It was originally cloned as a prostate-specific protein,but later it was also detected in lung cancer,breast cancer,colon cancer and cutaneum carcinoma.Recent studies have found that TRPM8 can not only sense cool stimulation,but also be used as a novel diagnostic marker and a therapeutic target for prostate cancer.This review focuses on the structure,function,localization and regulation of TRPM8 and its prospects in diagnosis and therapy of prostate cancer.
