鹿蹄草内生真菌的分离鉴定及其对鹿蹄草的生理效应 被引量:3

摘要 采用表面消毒法及选择培养基分离鹿蹄草根部内生真菌,通过初步回接试验筛选高效菌株,采用形态学及18SrDNA序列分析方法对高效菌株进行鉴定,并通过二次回接试验测定高效菌株对宿主的生理效应。结果表明,从鹿蹄草中分离到7株内生真菌,初步回接试验表明其对鹿蹄草的生长均有促进作用;筛选到一株高效菌株R25,鉴定结果表明该菌株为丛赤壳属(Nectria)真菌;该菌株能显著提高宿主根系活力,以及叶片中叶绿素a、可溶性糖和超氧化物歧化酶活性。筛选的高效菌株能显著提高鹿蹄草的生长代谢活性,在鹿蹄草的引种驯化过程中具有重要的应用价值。 Endophytic fungi were isolated from surface-disinfected samples by selective media of endophytic fungi, and high-effective strains capable of significantly promoting the growth of Pyrola calliantha H.Andr were screened by back inoculation experiment. They were identified by morphological and molecular methods. Influence of the high-effective strains on physiological effects of Pyrola calliantha H.Andr were also determined by further back inoculation experiment. The results showed that seven strains of endophytic fungi were isolated. All of them increased the survival rate of Pyrola calli- antha H.Andr, and the strain R25 significantly promoted the growth of the plant, which was determined as the efficient strain in this study, and was identified as Nectria. The strain significantly increased the root activity, Chla content, soluble sugar content, and the activities of SOD. The high-effective strain significantly promotes metabolism activity of the host, and has wide application prospect in the naturalization of Pyrola.
