摘要
在对紫杉醇和干扰紫杉醇测定的 6种常见紫杉烷进行色谱优化分离的基础上 ,建立了红豆杉细胞培养物中紫杉醇的高效液相色谱检测方法。样品经提取后 ,在KromasilC18柱上以乙腈 :水为流动相进行梯度洗脱 ,于2 2 7nm处进行检测。紫杉醇在 0 .2 μg/ml~ 2 0 μg/ml浓度范围内线性关系良好 ,经测定检测限为 0 .1μg/ml,检测精密度为 3.4% ,回收率为 88.4%
A HPLC method was developed for the determination of Taxol from callus cultures. Samples were extracted and separated on a Kromasil C18 column, and acetonitrile:water was used as the gradient elution mobile phase. Linear calibration graphs were obtained for Taxol over the concentration range of 0.2~20μg/ml. The limit of detection is 0.1 μg/ml(the signal-to-noise ratio is 2).The RSD is 3.4%,and the recovery of Taxol is 88.4%.
