摘要
目的探讨分析新疆哈萨克族食管癌患者食管癌组织及癌旁组织中金属硫蛋白3(metallothionein-3,MT-3)基因CpG岛的甲基化状态及其与食管癌发生的关系。方法采用甲基化特异性聚合酶链反应(MSP)的方法,检测33例哈萨克族食管癌患者食管癌组织和癌旁组织中金属硫蛋白3(MT-3)基因启动子区CpG岛的甲基化发生状况。结果 33例哈萨克族食管癌患者癌组织和癌旁组织标本中MT-3基因启动子区CpG岛甲基化的发生率分别为66.7%(22/33)和27.3%(9/33);癌组织与癌旁组织甲基化率差异有统计学意义(χ2=10.28,P=0.001)。结论 MT-3基因的甲基化可能与食管癌的发生与发展有关。
Objective To explore the relationship between the level of Methylation of the CpG island in the M T-3 gene in esophageal carcinoma in the Kazak .Methods The methylation of M T-3 gene was detected by methylation specific polymerase chain reaction (MSP) in 33 esophageal cancer tissues and adjacent tis-sues .Results T he methylation rates of the M T-3 gene in esophageal cancer tissues and adjacent tissues were 66 .7% and 27 .3% in Kazaks ,Significant differences were seen in the methylation rates of M T-3 gene in esophageal cancer tissues and adjacent tissues (χ2 =10 .28 ,P=0 .001) .Conclusion Aberrant promoter methylation of the CpG island in the M T-3 gene maybe play an important role in the pathogenesis of esoph-ageal squamous cell carcinoma in the Kazak .
