摘要
本文采用 PCR技术扩增和克隆了平胸龟的 Sox基因 ,并通过银染测序进行了序列分析。结果显示 ,平胸龟雌雄个体均能扩增出相同长度的片段 ,其 DNA序列与人 SRY基因的 HMG box同源性达 72 .7% ,推测的氨基酸序列与 SRY基因的 HMG box序列同源性为 4 6.5% ,充分显示出 Sox基因在系统进化上的保守性。本文为 Sox基因的起源和进化研究提供了资料。
The Sox gene of P. megacephalum were amplified and clond using PCR. The amplification fragment was sequenced by PCR silversequencing. The results indicated that the Sox gene fragment can be amplified in P. megacephalum of male and female. The homologous of Sox gene sequences comparing with HMGbox of SRY is 72.7% , and the sequence homologous of diving amino acid is 46.5%. This may be concluded that Sox gene is conservative in phylogenetic. This paper provide the date for Sox gene study of origin and evolution.
