摘要
在成熟神经系统中 ,嗅觉上皮 (OE)很特殊 ,它能不断产生新的神经元。本文用细胞培养、组织化学和免疫细胞化学技术对成年小鼠的OE进行了研究。实验显示 :双极细胞NF、NSE、MAP2、OMP和tau蛋白免疫染色为阳性 ,但keratin免疫染色为阴性 ,说明双极细胞是神经元。用不同浓度血清的培养基 ,离体培养成年小鼠的OE ,观察碱性成纤维细胞生长因子 (bFGF)和脑源神经营养因子 (BDNF)对OE细胞数量和突起发育的影响。结果显示 :将bFGF与BDNF的组合分别加到 10 %和 5 %血清的OE培养基 (OEM)中 ,增加了双极细胞的数量和比例 ,刺激了双极细胞突起的发育 ,减少了大细胞和梭形细胞。这种组合的效果比bFGF或BDNF单独作用好。在 10 %血清的OEM中 ,神经营养因子的效果比在 5 %血清的OEM中好。
Olfactory epithelium (OE) is unique in the adult nervous system for it produces new neurons throughout life. The cultured cells of adult mouse OE in vitro were studied by histochemistry and im^mun^ocyto^chemistry techniques, and the results showed that they were neurons as up to 50% of the bipolar cells were immuno^posi^tive to NF, NSE, MAP2, OMP and tau. The effects of basic fibroblast growth factor (bFGF) and brain derived neurotrophic factor (BDNF) in OE medium with different con^cen^tra^tion serum on OE cell number and development of cell processes were also observed. The results showed that the combination of bFGF and BDNF added to 10% and 5% serum OE medium increased the bipolar cell number and percentage, stimulated the process outgrowth of bipolar cells, and decreased the numbers of giant cells and fusiform cells. The effects of the combination of bFGF and BDNF in the 10% serum OE medium were more significant than those of bFGF and BDNF applied alone; and the effects of neurotrophic factors in the 10% serum OE medium were more significant than those observed in the 5% serum OE medium.
