摘要
用PAGE活性染色法分析了D.rndiodurans过氧化氢酶(Cat)和超氧化物歧化酶(SOD)._2种同种异型D.radiodurans(RI和Sark)的Cat在电泳带型上存在差异,两者Kat均可分为A、B和C3条带,但各带所占比例明显不同。SOD的分析结果表明,D.radioduransSOD以Fe2+和Mn2+离子的嵌合体形式存在,其中Fe-SOD成分占90%以上。PAGE活性染色法可检出Cat和SOD的最低菌体总蛋白量分别为1.2和2.0μg。
Catalase (Cat) and superoxide dismutase (SOD) of Deinococcus radiodurans were analyzed by polyacrylamide gel electrophoresis (PAG) combined with enzyme activity staining. It was found that electrophoresis pattern of Cats of D. radiodurans and D. radiodurans Sark, which are two kinds of strain type of the same species, differ from each other. Both strains' Cat display A, B and C bands. But the proportion of each band are coniderable different. Results of SOD analysis Of D. radiodurans indicated that SOD from D. radiodurans existed in the form of chimera with Fe2+ and Mn2+. Among them, the component of Fe - SOD occupied over 90%. PAG activity staining could check the ndnimum of total protein component per cell for Cat and SOD. They were 1. 2 and 2. 0μg respectively.
