紫红龙火龙果及其大果型芽变系的ISSR分析

Identification of Zihonglong Pitaya and Its Big-fruit Sport Line by ISSR

为揭示紫红龙火龙果及其大果型芽变系在DNA水平上的差异,采用ISSR分子标记技术对紫红龙火龙果及其大果型芽变系进行了遗传鉴定。结果表明:采用筛选出的9条引物,共扩增出67条重复性好的谱带,其中26条具有多态性,多态性比例为38.8%;应用引物811、835和834可以有效区分出紫红龙火龙果及其大果型芽变系,构建其DNA的ISSR指纹图谱;用遗传多样性分析软件NTSYSpc 2.10 e进行统计分析,得到供试材料间的遗传相似系数为0.6119。结果显示,紫红龙火龙果及其大果型芽变系在DNA分子水平上存在明显的遗传差异,大果型芽变系为紫红龙遗传上稳定的变异体,具有成为大果型新品种的遗传基础,可作为新型材料加以保护和利用。ISSR技术能有效地鉴定火龙果芽变。

To uncover the difference between Zihonglong pitaya and big-fruit sport line at DNA level, the genetic identification of Zihonglong pitaya and big-firuit sport line was conducted by using ISSR molecular markers. The results showed that by using the nine primers screened 67 reproducible bands were amplified, of which 26 were polymorphic, polymorphlc ratio was 38.8 % ~ primers 811,835 and 834 could of- fectively differentiate Zihonglong pltaya and its big-fruit sport line and construct the DNA ISSR fingerprint; the similarity coefl^clent among tested genetic materials was 0.6119 according to statistical analysis of genetic diversity by using NTSYSPO 2. 10 e analysis sct^are. Zihon- glong pitaya and its big-fruit sport line existed obvious genetic diversity at DNA molecular level, the big-fruit sport line was stable variants of Zihonglong pitaya with the genetic foundation d becoming a new big-fruit cuhivar and be protected and utilized as a new-type material. ISSR technology can be used to identify the pitaya bud variation effectively.
