摘要
用70%甲醇超声提取核桃仁中的总多酚,Folin-Ciocalteu比色法研究该提取物中总多酚含量测定的适宜条件。结果表明:核桃仁提取物加Folin-Ciocalteu试剂0.5 mL、10%Na2CO3溶液8 mL,反应温度25℃、反应时间2.5 h,在770 nm处测定吸光度,总多酚的含量与吸光度呈良好的线性关系。测定方法的平均回收率为98.4%,相对标准偏差(RSD)为4.28%。该方法具有操作方便,稳定性、重现性、精密度好等优点,适宜于核桃仁提取物总多酚含量的测定。
With 70 % methanol ultrasonic extraction of total polyphenols in walnut kemd, Folin-Ciocalteu colorimetry method to determine to- tal polyphenols in Walnut kernel was developed. The results showed that the reaction was carried out with 0.5 mL of Folin-Ciocalteu reagent and 8mL of 10 % aqueous Na2CO3 solution in temperature 2.5℃ and reaction time 2.5h. The detection wavelength was at 770 nm. Total polyphenols content and absorbency is good linear relationship. The average spiked recovery was 98.4 %, with a RSD of 4.28 %. This method is easy to operate with fast, sensitive, and stable determination of total polyphenols in mulberry fruits.
