摘要
目的对挤出法制备脂质体进行综述。方法参阅国外32篇文献,总结、归纳和分析了温度对挤出过程、挤出对粒径大小、双分子层数、渗透压、水及氨基酸渗透性和包封体积的影响。结果挤出法通过控制膜孔径使脂质体产生形变、破裂过膜,再重新组装闭合,从而得到粒径不同的新脂质体,当温度高于Tm时有利于这一过程的进行,同时挤出对脂质体结构以及小分子物质渗透性产生一定影响。结论挤出法具有工艺简便、重现性好、条件温和、所得制剂粒度均匀、包封体积大等优点,在脂质体的制备及工业化生产中具有很大的应用前景。
Objective To summarize the extrusion method of liposomes.Methods Referencing more than 30 overseas literatures,we concluded,analyzed and discussed the temperature′s effect on extrusion process,the influence of extrusion on particle size,number of bimolecular layers,the osmotic pressure,water and amino acids permeability and encapsulating volume.Results Extrusion method could make liposomes deformation and rupture,eventually pass through the membrane by controlling the membrane pore size,then package again,thus gave liposomes with various particle sizes.It was conducive to extrude when the temperature was higher than Tm,meanwhile extrusion had a certain impact on the structure of liposomes,as well as the permeability of small molecules.Conclusions Extrusion has many advantages,such as good reproducibility,mild conditions,uniform particle size,high encapsulated volume etc,also has great application prospects in the preparation of liposomes and industrial production.
