摘要
目的:通过检测转化生长因子βⅡ受体(TβRⅡ)在小鼠卵泡早期发育过程中的表达变化,为卵巢细胞间调控网络的研究提供理论基础。方法:取0、1、2、5、7天和11天雌性健康小鼠的卵巢组织。采用免疫组织化学法和实时荧光定量聚合酶链反应(Real-time PCR)检测卵巢组织中TβRⅡ蛋白及mRNA表达水平。结果:TβRⅡ蛋白在各组小鼠卵巢的卵母细胞胞浆中均有表达,原始生殖细胞囊中表达最强,始基卵泡中表达下降,初级、次级卵泡中表达增强。初级卵泡的颗粒细胞开始表达TβRⅡ,并随着卵泡发育表达逐渐增强。卵巢组织中TβRⅡmRNA表达水平随卵巢的生长发育先降低后增高,0、1天和11天小鼠卵巢组织中的TβRⅡmRNA表达水平约是2、5天和7天小鼠卵巢组织的2倍,差异显著(P<0.05)。结论:TβRⅡ在卵泡发育初始阶段的表达具有时间和空间的特异性,提示TGF-β信号通路在早期卵泡发育过程中可能起着重要的调节作用。
Objective:We provided a theoretical basis for the regulatory mechanism of follicular development by investigating the dynamic expressions of TβRⅡ proteins and mRNA during early folliculogenesis in mouse ovaries.Method:Ovaries were collected from the healthy neonatal female mice at the age of 0,1,2,5,7,and 11 days.The expressions of TβRⅡ proteins and mRNA were detected and evaluated by immunohistochemistry and Real-time polymerase chain reaction respectively.Results:TβRⅡ proteins were expressed in the cytoplasm of oocytes within ovaries in different age stages.The highest intensity of TβRⅡ staining was present in germ cell cysts.Primordial follicles exhibited moderate staining,and the staining of TβRⅡ increased during the transition to in primary and secondary follicles.For granulosa cells,TβRⅡ expression was found to be primarily in the primary follicles and increased with follicular development.There was no significant difference in TβRⅡ mRNA level among the ovary tissues of 0,1 and 11 days(P&gt;0.05).However,the ovarian TβRⅡ mRNA level during these periods was 2-fold higher than these of 2,5 and 7 days(P&lt;0.05).Conclusion:The expression of TβRⅡ proteins and mRNA in mice ovary suggests that the TGF-β signaling pathway may play an important role during early folliculogenesis.
