摘要
目的:探讨中药股密葆治疗大鼠激素性股骨头坏死的生物力学的作用机理。方法:2月龄清洁级SD大鼠180只,按体重根据随机数字表法,分为空白对照组、模型组、西药组(福善美组)、中药(高、中、低)剂量组6组,每组8只,雌雄各半。后5组参照S.Okazaki等应用大肠杆菌内毒素联合激素注射的造模方法构建大鼠激素性股骨头坏死模型。中药高、中、低剂量组分别灌服股密葆混悬液,西药组灌服福善美混悬液,空白对照组和模型组均灌服等量的生理盐水。分别于药物干预治疗的4、8、12周取大鼠右侧股骨头行生物力学实验检测,测量各组不同时间点软骨下骨最大轴向载荷、应力强度、最大位移值、轴向刚度;取左侧股骨头行病理组织学检查。结果:股密葆组的股骨头软骨下骨的最大轴向载荷、应力强度、位移及轴向刚度,与模型组相比较差异有统计学意义(P<0.01)。股密葆组治疗激素性股骨头坏死随着治疗时间的增加,疗效更加明显。病理学结果显示,股密葆组的股骨头软骨下骨的骨小梁排列较整齐,骨细胞凋亡较少,髓腔内血栓形成较少。结论:中药股密葆能显著改善激素性股骨头坏死大鼠骨结构力学及材料力学性能,提高骨质量。
Objective:To investigate the biomechanic effect of Gumibao on steroid-induced osteonecrosis of the femoral head (SINFH) in rats. Methods: One hundred and eighty 2-month-old SD rats were randomized into six groups, blank control group, model group, Fosamax group, Gumibao groups (High, medium and low-dose groups), and 8 rats in each group. SINFH models were established according to S. Okazaki's technique. Rats in Gumibao groups received intragastric administration of Gumibao suspension. Fosamax group was given Fosamax. Axial loading, stress, displacement and axial stiffness of the right femoral heads were measured at week 4, 8 and 12 after treatment. At the same time, the left femoral heads were subjected to observation of pathological features. Results:The axial loading, stress, displacement and axial stiffness of the Gumibao groups were much greater than those of model group (P〈0.01). In the pathological studies, the trabecula of the femoral heads of Gumibao groups arranged in order, with less apoptosis in osteocyte and less thrombus in the bone marrow compared with the model group. Conclusion:Gumibao could significantly improve the bone structural mechanics and material mechanical properties and the quality of bone in rats with SINFH.
