摘要
目的:研究miR-126在肺癌中与血管内皮生长因子(VEGF)蛋白表达的相关性,探讨miR-126与肺癌的作用机制。方法:采用组织芯片平台,应用原位杂交方法(IsHH)检测56例肺癌组织、41例相应的癌旁组织和15例正常肺组织的miR-126表达;免疫蛋白印迹法(WestemBlotting)检测VEGF蛋白的表达。结果:肺癌组织miR-126阳性表达率均显著低于癌旁组织和正常肺组织(P〈0.05);vEGF蛋白在肺癌组织中的表达率均显著高于癌旁组织和正常肺组织(P〈0.05)。肺癌组织中miR-126的低表达与肿瘤的淋巴转移密切相关,与病理分级有一定关系。相关性检验表明miR.126与VEGF表达呈显著负相关(P〈0.05)。结论:miR-126可能作为肺癌重要的抑癌基因及其转移的潜在标志。
Objective: To study the expression correlation in miR-126 and protein of vascular endothelial growth factor (VEGF) within lung cancer, and to explore the action mechanisms of miR-126 and lung cancer. Methods: Using tissue microarray platform, miR-126 expressed in lung cance: tis- sues of 56 cases and corresponding paracancerous tissues of 41 cases, and normal lung tissue of 15 cases were examined, applied with situ hybridiza- tion (ISHH); to detect the expression of VEGF protein with western blotting. Results: The positive expression rate of lung cancer with miR-126 was significantly lower than that of paracancerous tissues and normal lung tissues (P〈0.05); the expression rate of VEGF protein in lung cancer tissues was significantly higher than that in paracancerous tissues and normal lung tissiaes (P〈0.05). The low expression of miR-126 in lung cancer tissues was closely related with lymphatic metastasis of tumor, where was a certain relationship with the pathological grading. Correlation tests showed that miR-126 and VEGF expression were significantly negative correlation (P〈0.05). Conclusion: miR-126 can be used as an important tumor suppressor gene of lung cancer, and potential mark of its metastasis.
