摘要
本文研究青梅核醇提物乙酸乙酯相(PMS-E)和青梅花醇提物正丁醇相(PF-B)对小鼠高尿酸血症的拮抗效应。采用酵母膏灌胃建立小鼠高尿酸血症模型,碱性磷钨酸盐法测定小鼠血尿酸水平,试剂盒法测定小鼠血清肌酐、尿素氮和白球比水平以及血清和肝脏中黄嘌呤氧化酶(XO)活性。结果表明,PMS-E和PF-B均能显著降低高尿酸血症小鼠血尿酸水平(p<0.05)和白球比,并能显著升高血清肌酐(p<0.05)和尿素氮水平(p<0.05);并且在一定程度上促进动物的体重增长,而对肝体指数和肾体指数均无显著影响。此外,PMS-E和PF-B可有效抑制血清和肝脏XO活性。本研究结果为用PMS-E和PF-B开发降尿酸、抗痛风的膳食补充剂奠定了基础。
The antagonism effects of ethyl acetate fraction of Prunus mume seed extract (PMS-E) and n-butanol fraction of Prunus mume flower extract(PF-B) against yeast extract induced hyperuricemia in mice were investigated. The mice hyperuricemic model was made .by orally administering yeast extract; serum uric acid level was measured by alka- line phosphotungstate method; creatinine, urea nitrogen and albumin and globulin ratio in serum, and xanthine oxidase (XO) in serum and liver were measured by corresponding kit. The results indicated that PMS-E and PF-B could reduce the serum uric acid level and albumin and globulin ratio in hyperuricemic mice, and increase the serum creatinine, urea nitrogen levels significantly. Moreover, PMS-E and PF-B could increase body weight of hyperuricemic mice, but has no significant influence on liver-somatic index and kidney-somatic index. In addition, PMS-E and PF-B could inhibit XO activity in serum and liver significantly. This study laid a foundation for PMS-E and PF-B exploitation to dietary supple- ments for lowering uric acid and anti-gout.
