摘要
目的观察川芎嗪对人肝癌HepG2细胞的增殖抑制和诱导凋亡作用及其与突变型p53蛋白表达变化的关系。方法应用不同浓度的川芎嗪作用于人肝癌HepG2细胞24h、48h和72h,MTT法检测细胞活力;Hoechst染色法检测细胞凋亡;免疫细胞化学法检测川芎嗪对突变型p53蛋白表达的影响。结果不同浓度的川芎嗪呈时间、剂量依赖性地抑制人肝癌HepG2细胞的增殖并促进其凋亡(均P<0.01),800mg/L川芎嗪作用细胞72h后,其抑制率及凋亡率分别为49.59%和31.31%。免疫细胞化学法显示,不同浓度的川芎嗪作用人肝癌HepG2细胞48h后,突变型p53表达呈显著降低(均P<0.01)。结论川芎嗪可抑制人肝癌HepG2细胞的增殖,促进细胞凋亡,其机制可能与其抑制突变型p53蛋白的表达有关。
Objective The effect of Tetramethypyrazine ( TMP ) on the proliferation , apoptosis of the human liver cancer HepG2 cell and its relationship with the expression of mutant p53 ( mt p53 ) were studied.Methods HepG2 cells were treated by different concentrations of TMP for 24 , 48 and 72h.The cells′proliferative vitality was detected by MTT assay , and the apoptosis of HepG2 cells was detected by Hoechst stain.After TMP at different concentrations was added to the media respectively for 48h , the expression of mt p53 protein was detected by immunocytochemical staining.Results TMP at different concentrations in our study could inhibit the proliferation and induce apoptosis of HepG2 cancer cells in a dose-and time-dependent manner ( both P&lt;0.01 ), the inhibition rate and apoptosis rate reached 49.59% 、31.31% after treatment with 800mg/L TMP for 72h.After HepG2 cells were treated by different concentrations of TMP for 48h , mt p53 protein expression was significantly down-regulated in a dose-dependent manner ( both P&lt;0.01 ) .Conclusions TMP can inhibit the proliferation and induce the apoptosis of the human liver cancer HepG2 cell , which may be related to the down regulation of mt p53 protein expression.
