白酒对大鼠心肌凋亡基因bcl-2、caspas-3表达水平的影响

摘要 目的观察不同剂量白酒对大鼠凋亡基因bcl-2、Caspas-3表达的影响。方法140只2~3月龄健康SD大鼠随机分成7组:正常对照组及不同剂量白酒灌胃模型A组(0.8mL/kg.d)、B组(1.6mL/kg.d)、C组(2.4mL/kg.d)、D组(3.2mL/kg.d)、E组(4.0mL/kg.d)和F组(4.8mL/kg.d),每组20只。对照组每日给予10mL/kg.d生理盐水灌胃,其余模型组分别给予53度白酒灌胃,每周灌胃6次,称重一次,连续灌胃16周。16周后处死大鼠取心肌组织进行病理学检查,同时取大鼠心肌组织用TUNEL法检测细胞凋亡相关蛋白bcl-2、caspase-3的表达水平。结果bcl-2、caspase-3在对照组、模型A、B、C组心肌细胞浆中呈低水平表达,差异无统计学意义(P>0.05);模型D、E、F组与对照组比较bcl-2、caspase-3表达水平明显升高,差异有统计学意义(P<0.05);模型D、E、F组与A、B、C组相比bcl-2、caspase-3水平明显升高,差异有统计学意义(P<0.05)。结论适量白酒不会引起大鼠凋亡基因bcl-2、Caspas-3表达变化,但过量白酒(大于3.2mL/kg.d)摄入可导致bcl-2、caspase-3表达上调,加速心肌细胞的凋亡和坏死。 Objective Different doses of liquor rat apoptotic gene BCL-2,caspase-3 expression was observed. Methods 2 to 3 months, a total of 140 healthy SD rats were randomly divided into 7 groups which were normal control group and different doses of alcohol toxicity model group were group A (0.8mL/kg. d), groupB (1.6mL/kg. d), group C (2.4mL/kg · d), group D (3.2mL/kg · d), group E (4. 0mL/kg· d), and group F (4. 8 mL/kg ·d), (n=20). The control group to give 10mL/kg · d saline, and the rest of the model group were given 53 degrees of alcohol toxicity, weighing once a week, gavage 6 times each week and intragastrically continual for 16 weeks. The rats were sacrificed myocardial tissue for pathological examination after 16 weeks. At the same time took the rat myocardium apoptosis-related proteins bCL-2, caspase-3 expression levels detected by TUNEL method. Results bCL-2, caspase-3 in the control group, models A, B, C myocardial cytoplasm was expressed at low levels, and there were no statistically significant differences (P〉0.05). Model D, E, F group and control group comparison of bCL-2, caspase-3 expression levels were significantly increased, and there were statistically significant differences (P〈0.05). Model D, E, F compared with group A, B, and C, the expression of bCL-2, caspase-3 level was significantly higher, and there were statistically significant differences (P〈0.05). Conclusion The right amount of liquor will not cause the rat apoptotic gene bCL-2,caspase-3 expression changes, but excessive the liquor (greater than 3.2mL/ kg · d) intake can lead to the expression of bCL-2, caspase-3 expression, accelerated myocardial cells apoptosis and necrosis.
