摘要
目的探讨京尼平(genipin)对肾癌是否有治疗作用。方法将0μmol/L、40μmol/L、80μmol/L和120μmol/L四种浓度的genipin溶液分别浸染肾癌细胞48h,MTT法检测细胞的增殖情况;聚合酶链反应检测UCP2的基因表达。使用荧光探针检测细胞活性氧分子(ROS)含量。结果genipin在中、高剂量组能明显抑制肾癌细胞的增殖,随着给药浓度的增加,细胞增值抑制作用逐渐增高(P<0.005)。genipin能抑制肾癌细胞中UCP2的表达,在中、高剂量给药组,UCP2的基因及蛋白的表达显著抑制(P<0.05)。genipin能提高细胞内ROS的含量,在中、高剂量给药组ROS含量均显著提高(P<0.05)。结论genipin能明显抑制肾癌细胞的增殖,促进其凋亡,UCP2可能参与其作用机制。
Objective To investigate whether there is any therapeutic effect of genipin to renal cell carcinoma. Methods Renal carcinoma cells were treated with different concentrations(0 μmol/L, 40 μmol/L, 80 μmol/L, 120 9mol/L)of genipin. Cell growth inhibition was detected by assay of MTT. Expression of UCPz was detected by PCR methods. Contents of ROS were tested by fluorescence probe. Results Genipin significantly suppressed the growth rate to renal carcinoma cells, statistical significance were found in 80 /xmol/L and 120μmol/L groups. Expression of UCP2 was significantly inhibited in 80μmol/L and 120 μmol/L groups and there were statistically significant differences (P〈 0.05). Genipin can improve contents of ROS and was founded significantly increases in 80μmol/L and 120ptmol/L groups. Conclusion Genipin can obviously inhibit renal carcinoma cells growth and promote the apoptosis, and UCP2 may be involved in its mechanism.
