摘要 本工作对 188Re直接标记抗人结肠癌单克隆抗体 CL- 3的方法进行了详细的研究。以抗坏血酸作为抗体的还原剂 ,以 Sn Cl2 -柠檬酸还原 188Re O4 -溶液 ,进行抗体的直接标记。本方法操作简单、方便 ;标记率高 ( >95% )胶体含量小 ( <5% ) ;标记完后不需纯化分离 ;标记物有良好的体外稳定性和较高的免疫活性 。 The direct labeling method of monoclonal antibodies with generator produced rhenium 188 is studied in detail. Ascorbic acid is used as the reducer of antibodies and the protective agent against air oxidation as well. 188 ReO 4 - is reduced with SnCl 2 in the presence of citrate, which also acted as an intermediate complexing agent in the subsequent labeling step. The whole procedure is optimized and could be completed with 3 h. The final product possesses the radiochemical purity of higher than 95% with colloidal Re Less than 5%, and good stability in vitro as well as higher immunity.
