重组人生长激素对梗阻性黄疸大鼠肠黏膜细胞间黏附分子-1的影响 被引量:1

摘要 [目的]探讨重组人生长激素对梗阻性黄疸大鼠肠黏膜细胞间黏附分子-1(ICAM-1)的影响.[方法]取雄性Wistar大鼠随机分为假手术组、梗阻性黄疸组及重组人生长激素组,每组再分为7,14d组.各组均无菌操作,假手术组仅游离胆总管,梗阻性黄疸组和重组人生长激素组均双重结扎并切断胆总管.重组人生长激素组于手术当日始,每日在双后肢内侧轮流皮下注射给予0.75U/kg重组人生长激素.分别于术后7,14d采集门静脉血,并取回肠末端小肠组织;检测总胆红素、直接胆红素、丙氨酸转氨酶(ALT)和血浆内毒素.小肠组织行HE染色后观察黏膜病理形态学改变,用免疫组织化学SP法检测及Image Pro Plus软件图像分析手段判定黏膜表面ICAM-1水平.[结果]随着梗阻时间的延长,梗阻性黄疸组和重组人生长激素组大鼠血清总胆红素、直接胆红素和ALT值均升高,但重组人生长激素组较梗阻性黄疸组升高幅度小.梗阻性黄疸组和重组人生长激素组肠黏膜损伤程度和内毒素水平随着梗阻时间的延长均有不同程度的加重和升高,但重组人生长激素组损伤程度较梗阻性黄疸组轻.梗阻性黄疸组和重组人生长激素组肠黏膜ICAM-1的表达水平均高于假手术组,但重组人生长激素组低于梗阻性黄疸组,且随着梗阻时间的延长ICAM-1的表达水平略降低.[结论]外源性生长激素可降低梗阻性黄疸大鼠肠黏膜ICAM-1的表达,并可维护肠黏膜的屏障功能,降低血浆内毒素水平. OBJECTIVE To study the effects of recombinant human growth hormone (rhGH) on ICAM-1 of intestinal mucosa in rats with obstructive jaundice (OJ). METHODS Wistar rats were randomly divided into sham operation (SO) , 0J and rhGH groups, and in each group, rats were randomly assigned into 7 and 14 days groups. The rats in SO group were only undergone laparotomy with dissociation of the common bile duct (CBD) , and in OJ and rhGH groups were undergone laparotomy with ligation and transection of the CBD. The rats in rhGH groups were performed subcutaneous inject ion with 0.75 U/kg per day of rhGH to posterior limb wall. On postoperative 7 and 14 days, the blood from the portal vein and the tissues of terminal ileum were taken to measure the TB, DB, ALT and endotoxin, and to observe the morphological changes and content of ICAM-1 by HE staining and immunohistochemistry with Image Pro Plus. RESULTS With the time of BDL extending, the values of serum DB, TB and ALT were increased in rhGH and OJ groups, but lower in rhGH group than in OJ group, and the degree of ileal mucosa injury and the levels of endotoxin in OJ and rhGH groups were increased, but slight higher in rhGH group than in OJ group. The levels o f ICAM-1 were more higher in OJ and rhGH groups than in SO group, but lower in rhGH group than intestinal mucosa in endotoxin of blood in OJ rats group.
