5′端非翻译区和NS5B区在丙型肝炎病毒基因分型中意义的再评价 被引量:1

摘要 目的研究丙型肝炎病毒(HCV)的5′端非翻译区(5′-UTR)和NS5B区对中国人群感染的HCV基因分型的确切意义。方法选择NCBI核苷酸数据库中来源于中国人的HCV全基因组,利用NCBI基因分型工具,对同一条HCV序列中5′-UTR区、NS5B区和全长基因组进行基因分型,确定5′-UTR区和NS5B区在HCV基因分型中的全基因组代表性。在此基础上,选择NS5B区对125例临床HCV-RNA大于1.0E3的标本进行了基因分型。结果与全长基因组的分型结果相比,5′-UTR区基因分型在基因型判断上存在差异,仅有45%的符合率,特别是其无法精确判断基因亚型;而NS5B区基因分型结果与全长基因组的分型结果无论在基因型还是在基因亚型的判断上都具有良好的一致性,符合率可达95%;利用NS5B区对临床125例标本成功进行了基因分型,结果显示以1b和2a两种亚型为主。结论 NS5B区在HCV基因分型中具有良好的全基因组序列的代表性,能够有效用于临床常规的HCV基因分型检测。
