摘要
目的 探讨白细胞介素-27(IL-27)在强直性脊柱炎(AS)患者外周血的表达及其对CD4+T细胞的增殖分化作用机制.方法 分别收集AS患者、健康体检者(Health controls,HCs)的外周血,磁珠分离CD4+T细胞.ELISA测定IL-27及其相关细胞因子的表达.IL-27体外作用后,CCK-8测定细胞增殖情况,定量PCR分析T-bet和GATA3基因表达情况.结果 AS患者外周血IL-27显著高于HCs组(P<0.01),IL-27作用后,AS组和HCs组CD4+T细胞增殖能力均增强,AS组CD4+T细胞增殖能力强于HCs组(t=12.1,P<0.01),同时AS组细胞培养液中IFN-γ在IL-27作用后较HCs组均显著增高(P<0.01),IL-2及IL-10表达无明显变化.IL-27诱导后AS组CD4+T淋巴细胞T-bet基因表达显著增加(t=14.3,P<0.01),而GATA3差异无统计学意义.结论 IL-27在AS患者外周血表达升高,并可诱导CD4+T细胞增殖,可能通过上调T-bet的表达诱导CD4+T细胞向Th1分化,可能在AS免疫炎性反应过程中发挥重要作用.
Objective To investigate the role of interleukin-27 (IL-27) in proliferation and differentiation of CD4+ T cells in ankylosing spondylitis(AS). Methods CD4+T ceils were separated from peripheral blood which col- lected from AS patients and health controls (HCs). Cells proliferation was detected by CCK-8 kit, and cytokines were analyzed by ELISA. Real-time PCR was used to determine the mRNA expression of T-bet and GATA3. Results Serum IL-27 level in patients with AS was obviously higher than that in HCs(P 〈0.01 ). The proliferation rate of CD4+ T ceils and the level of IFN-γ in cultured medium in AS were higher than those in HCs group after IL-27 stimulation (P 〈0.01 ). IL-27 could induce T-bet mRNA expression in CD4+T cells in AS (t = 14.3 ,P 〈0.01 ) ,but no change was found in GATA3 mRNA expression. Conclusions IL-27 could induce the proliferation of CD4+ T cells and the activation of T-bet pathway in AS. Furthermore,Thl immune response and related cytokines could be induced by IL- 27 in AS. These implicate that IL-27 may play an important role in AS related inflammation.
