摘要
目的:探讨原花青素(PC)对氨基脲(SEM)染毒小鼠睾丸组织形态及睾丸酶损伤的修复作用。方法:50只昆明种小鼠按体重随机分为5组:溶剂对照组、SEM染毒组、SEM染毒+低、中、高剂量PC保护组。SEM染毒组小鼠按56.25 mg/(kg.bw)剂量灌胃SEM溶液;低、中、高剂量PC保护组小鼠分别按100、200、400mg/(kg.bw)剂量灌胃PC溶液。除溶剂对照组灌胃等体积的去离子水外,其它每组均上午灌胃SEM、下午灌胃PC,每天1次,实验时间均为6周。实验结束后处死动物,分离双侧睾丸测定相关指标。结果:溶剂组,睾丸组织形态正常;SEM染毒组睾丸曲细精管上皮细胞层次显著减少,管壁脱落缺失,腔内未见精子等;PC保护组睾丸形态较SEM染毒组有所改善,且随PC剂量的增加改善较明显,睾丸间质有所缩小,曲细精管上皮细胞层次趋向正常等。SEM染毒组小鼠睾丸组织LDH-X、γ-GT活性降低,ACP活性升高(P<0.05);中、高剂量PC保护组LDH-X、γ-GT活性升高,ACP活性降低(P<0.05);SEM染毒组睾丸组织GSH-PX活力及GSH含量下降(P<0.05);高剂量PC保护组GSH-PX活力、GSH含量均呈增高趋势,差异均有统计学意义(P<0.05)。结论:56.25 mg/(kg.bw)剂量SEM对小鼠睾丸组织形态及其酶有损伤作用;一定剂量的PC对SEM损伤的睾丸组织形态及其酶有修复作用,其作用机制可能与抗氧化损伤有关。
Objective: To investigate the repairment effect of procyanidins(PC) on damaged testicular tissue in semicarbazide(SEM) - exposed mice. Methods: 50 male Kunming (KM) mice were randomly divided into five groups depending on weight: solvent control group, SEM exposure group, SEM exposure+ low, medium and high doses of PC protection group. The mice in solvent control group were intragastric deionized water once a day for 6 weeks. Mice in SEM exposure group were intragastric 56.25 mg/(kg.bw)SEM solution. Mice in low, medium and high doses of PC protection groups, were intragastic 56.25 mg/(kg. bw) SEM solution in the morning, and were intragastic 100mg, 200 mg, 400 mg/(kg. bw) PC solution respectively in the afternoon for 6 weeks. After experiment, mice were taken blood and killed, seperated bilateral testes and the correlated detection indicators were measured. Results: The histomorphology of mice testicular tissue in solvent group was normal, and in SEM exposure group was changed with epithelium layer of testicular contorted seminiferous tubules reduced significantly, the tubal wall defluxion missing, and without intraluminal sperm, etc. Testicular morphology of mice in PC protection group has improved compared with SEM exposure group, and was related with the dose of PC obviously. These improvement were interstitial tissue of testis minification, epithelium layer of testicular contorted seminiferous tubules tending to normal, etc. The activity of LDH - X, γ - GT and GSH- PX and the level of GSH in testicular tissue of mice in SEM exposure group were decreased and activity of ACP was increased( P 〈 0.05 ). However, the activity of LDH- X, γ - GT in testicular tissue of mice in medium and high doses of PC protection group were increased and activity of ACP was decreased( P 〈 0.05), meanwhile the activity of GSH- PX and the level of GSH in testicuIar tissue of mice in high doses of PC protection group were increased, the difference had statistical significance ( P 〈 0.05 ). Conclusion: There was damage effect of 56.25 mg/( kg. bw) dose SEM on morphology of mice teatieular tissue and enzyme. A dose of PC have repair effect on morphology and enzyme of testicular tissue iniuredby SEM. The mechanism of PC may be associated with resistance to oxidative damage.
