力竭运动大鼠心肌心钠素表达的变化 被引量:41

摘要 为了探讨力竭运动对大鼠心脏心钠素表达的影响 ,采用 4月龄雄性 SD大鼠 42只 ,体重 2 2 0 g~ 36 0 g,随机分为 1正常对照组 (A组 )、2有训练游泳力竭组 (B组 )、3无训练游泳力竭组 (C组 )。每组各 14只。灌注后取出整心 ,用免疫组化 ABC法和图像分析技术以及透射电镜实验 ,显示和观察大鼠心肌心钠素的表达和心钠素电子致密颗粒的变化。结果是 :心钠素免疫阳性反应为 :右心耳 >右心房 >左心房 >右心室近心房处。与 A组相比 ,B组大鼠心房肌心钠素阳性反应明显增强 ,心钠素电子致密颗粒大量增加 ;C组大鼠心房肌心钠素表达明显减少 ,仅右心房肌细胞有少量心钠素颗粒。两组大鼠心肌组织和细胞超微结构出现程度不同的损伤性变化 ,以 C组大鼠心肌细胞结构的损伤性改变更明显。结果表明 :力竭运动可对大鼠心肌细胞的结构和心钠素表达产生损伤性影响。 The effects of exhaustive exercise on the expression of atrial natriuretic factor (ANF) in cardiac myocytes is studied. 42 4 months old SD male rats, weight 220 360g, were randomly divided into groups of sedentary control (group A, 14), exhaustive swimming with suitable training in advance (group B, 14) and only exhaustive swimming (group C, 14) and the whole hearts were removed from all rats. The expression and electron dense granules of ANF in cardiac myocytes were investigated by immunohistochemical reaction, image analysis and transmission electron microscopy (TEM) experiment. The results showed a right auricle > right atrium > left atrium > the part of right ventricle near the atrium in ANF expression of cardiac myocytes. The immunohistochemical positive reaction and electron dense granules of ANF in left and right atrium myocytes were all obvious increased in group B and significantly decreased in group C, separately, compared with group A (p<0.01). The histological appearance and ultrastructure of the cardiac myocytes were lightly injured in group B and in seriously abnormal conditions in group C. The results demonstrate that the structures and ANF expression of cardiac myocytes of rats may be terribly affected by only exhaustive exercise training.
