摘要
目的探索人结肠癌细胞SW480转染4IgB7-H3后对糖基转移酶133GnT8和细胞表面多聚乳糖胺糖链表达的影响。方法脂质体介导的重组表达载体plRES2-EGFP/4IgB7-H3转染SW480细胞,筛选并获得稳定表达4IgB7-H3的亚克隆细胞株;RT-PCR、WesternBlot检测B3GnT8的表达;流式细胞术检测多聚乳糖胺寡糖链的表达。结果成功获得稳定表达4IgB7-H3的亚克隆细胞株SW480/4IgB7-H3;转染4IgB7-H3的SW480细胞中B3GnT8和多聚乳糖胺糖链的表达均明显下调(P〈0.05)。结论人肠癌细胞中4IgB7-H3过表达能抑制p3GnT8的表达,进而抑制多聚乳糖胺糖链的表达。
Objective To explore the expressions of glycosyltransferase ]3 3GnT8 and polylactosamine on cell surface in human colon cancer cell line SW480 transfected with Flow cytometry detcted lactosamine oligosaccharide chain expression.Results 4IgB7-H3 recombinant expression vector was successfully constructed.Then stably transfected SW480 cell line was got.Both the expressions of [3 3GnT8 and poly lactose amine sugar chain decreased significantly(P 〈 0.05) after transfection.Conelusi0a 4IgB7-H3 overexpression in human colorectal cancer cells can inhibit [3 3GnT8 production,and thereby inhibit poly galoctosamine extension.
